We are in Venice and leave for the US Friday , May 23 at 1:20. We are staying in the San Polo district near the Rialto Bridge. How is the best way to get to the airport and how long will it take? We have 2 carry ons and 2 medium back packs. In the 5 weeks we've been in Italy, I've been able to ask locals for information. Not having much luck this time. Thanks

I presume you leave at 13:20 (1:20pm), because at 1:20am there is only the water taxi option, Alilaguna doesn't run that early.
Besides the Alilaguna boats given above, you could take a regular vaporetto to Piazzale Roma then from there you can take the regular bus to the airport. That is probably the cheapest option.
The shuttle bus from Piazzale Roma to VCE is the no.5 AEROBUS of the ACTV city transit agency.

I have found the vaporetto and airport bus combo to be quite easy to manage (including with slow-moving parents). If you have a vaporetto pass, it is also good on the ACTV-brand airport bus (but not ATVO).
Having just purchased a vaporetto pass today I am fairly sure it does not include the ACTV bus to the airport, that is an additional ticket, I believe 4 euros with the pass and 6 without.
You can board the Alilaguna at Rialto; just make sure you find the right dock as there are several. It is not the one closest to the bridge but farther down. If you want to buy tickets in advance for the Alilaguna you go to the travel agent in the campo nearby ---I will have to look up the name for you. The Alilaguna goes to the airport across the lagoon. It is a couple of euros more than the vaporetto plus bus combination but you do not have to make a transfer to a bus.
Agree the vaporetto and bus combo is easy. We were a family of 6 (3 generations), each with carryon and backpack. Our flight left at 7:30am and we used the vaporetto and bus with no problems, even at the early hour we left our hotel. We bought the 72 hour vaporetto pass without the additional 4Euro for the bus to the airport and assumed our pass would not work on the bus. Our B&B told us that they thought the pass was also good for the bus so we tried it and it did work. Not sure why but it saved us the cost of the bus and made our pass even more worth while.
