This route from Google Maps takes you right through San Gimignano. That is a great spot to visit on the way to Montepulciano.
You could drive south on the historic RT.222, Via Chiantigianna, then cut over to the east to Montepulciano after seeing Siena.
Along 222, you should stop in Greve, Panzano, Castellina in Chianti, Radda in Chianti and, of course, Siena. These are towns with charm, character and history set amidst vineyards.

I agree. The link to the other route takes you down a main highway, which will get you from one place to the other, but is not nearly as scenic as 222.
Ditto for the SR222 route ! Lovely winding road and the towns suggested are delightful.
Stop whenever you see a village of interest !
I also agree with SR222. Much more scenic and goes through more wonderful towns. Stop in Panzano In Chianti and visit the singing butcher.
