Coming from California, which airport would be the best choice to fly into, Bari or Brindisi?

They are not particularly far apart, and you've got to come a long way--whichever one offers a good schedule and price will work just fine. (Check Lufthansa)
Brindisi is much closer to Lecce, but if Bari works better, plan a brief stop along the coast, in the Itria Valley, or in Matera before heading down to Lecce.
We were in Puglia in May. Bari to Lecce is a fast train ride with very frequent departures. We flew out if Bari. While a small point the Bari airport is quite pleasant!
Coming from California, Bari is closer.
BRI is also bigger, with more flights.
Check both and whatever works for you, that is the better airport.
I am in Lecce now and have been for 4 days. It is without doubt one of the jewels of Italy and a fantastic city. I Have been here in the Salento for a bit more than a month. The food is great as are the people and the wine and the scenery. You will love it I am sure. We stayed almost a full month in a great AirBnb in Marina di Novaglie. It worked out to be about $35 per day for a two bedroom house fully stocked with a magnificent view out over the Adriatic.
But for your trip: There are fast trains between Bari and Lecce...a Frecciargento... and this is a great way to get there. But there are direct fast Freccia trains from Rome to Lecce also about 5 hours. I don’t know your flight details or how much the cost is to get to Bari or how many changes you have but the train is another option at about 104 Euro for TWO people.
