I will be driving from Zurich to the Dolomites in northern Italy. I plan to cross into Austria near Diepoldsau Sz. and will be on Austrian Hwy E60 after I cross the border. What is the best way to obtain the Austrian Motorway Vignette for my trip?
The vignette can be bought online (if you know your plate number), it is often available at service stations near the border; if everything fails, in Austria is sold also at post offices and tobacconists.
Remember that the Arlberg highway tunnel this year is closed for maintenance from April 15th to November 22nd.

I got mine literally, just after crossing the border.
you will know your number plate as soon as you drive off the lot. I takes about 5 seconds - in English, on line. No fuss, no muss. Especially if you open it and do a dummy run first.
just an FYI, my experience getting an Austrian Motorway Vignet was not good. We crossed the border near Oberriet and had a tough time finding one. The post office in Austria did not sell them and we had to stop at 6 gas stations before we found one.

Did you have to cross the border right after getting the car in Zürich? I acknowledge that getting a rental means a few minutes probably on wifi to do it and put in the rental's number plate number.
