I plan on traveling from Milan to Genoa on the 15th of August by train. I have seen that the region celebrates the Assumption of Mary in the country. Although in the US it is a holy day of obligation for Catholics from what I have read the day in Italy is celebrated with closures, festivities, fireworks and often a 3 day weekend for Italians. I was hoping I could get some information on what to expect during the holiday and if trains run during this period. Thanks!

Trains from Milan to Genoa (or anywhere else) will run regularly.
They might be a bit crowded with people heading to the beaches of Liguria, but they will run regularly. If you leave in the morning with a high speed Intercity or FrecciaBianca train, it might be a good idea to book in advance. If you take Regionale Veloce (RV) trains, then there is no need, as they are first come first served. I would go with the RV trains. They take just 20 min longer and they are 7€ cheaper.
In that period Italians head to the beach towns, or lake towns, or mountain resort towns, all of which will be packed with visitors and where everything will be open for the visitors (shops, restaurants, entertainment, etc).
The cities, like Milan and also Genoa, will be emptier (Italians skip town on that weekend) with many stores and businesses closed. However you will be able to still find plenty of restaurants open, especially in the city centers. Museums should be open, but you need to check the opening times in the individual venues' websites. I don't know if they have special events in Genoa, or surrounding towns, for that weekend. If you go to popular seaside destinations near Genoa (like Camogli, Santa Margherita, Paraggi, Portofino, etc.) they will almost certainly be packed with people.
