We've arrived in Assisi for 2 nights, going to Siena on Monday. Was planning to take Flixbus, but just checked and there is only 1 bus that day and it is sold out. Looking for other options- most likely the train (either to Florence and then bus to Siena, or other train more direct). Was looking for car rentals, don't seem to be any in Assisi. Maybe there is some Tuscany tour one way to Siena??? We are pretty flexible, open to exploring areas on the way, although we will have backpack luggage.

We found we can get to Perugia, a short train from Assisi. From there, 1:20 bus ride to Siena. A lot less travel time!
There is a direct (no connection) train from Assisi to Perugia (30m). The train from Perugia to Siena requires a transfer taking 4h. What date are you using?
Bus from Assisi to Perugia is fast and easy --- we did the round trip in 2021.

The bus ride was a bit pricey + Flixbus charges a transaction fee. Oh well
