Assisi to Ostuni

I have three days to drive down to Ostuni from Assisi. I have seen Matera, Rome and Montipuciano - any other suggestions on where to stop - it could be three different places, one night in each, or three nights in one location.

Caveat: I have never driven in Europe.

It appears that driving straight through will take the better part of a day by the time you stop for a meal, so you have two extra days to play with.

What time of year? If you're talking summer, I'd consider stopping for at least a day on the Gargano Peninsula. I think the temperature would be more pleasant in the interior than it would be farther south. If at other times of the year, I'd be tempted to use the extra time in Puglia, which has lots of attractive towns. Alberobello is probably the most well known, but Bari (larger), Martina Franca, Monopoli, Locorotondo and Polignano a Mare are also good destinations, and that's just the places I know of quite near your driving route. I like Lecce a lot, but that would mean driving a bit beyond Ostuni.

If you are planning extra time in Ostuni for day trips, most of those places in Puglia will be within reach, in which case it would make more sense to spend the extra time farther north. Up near Assisi you have Spello and Spoleto, both of which other people have liked. I haven't seen either one. Spoleto would mean driving south for perhaps 30 miles before turning east, adding perhaps an hour to your driving time.
