My husband and I will be visiting Rome in October. There are conflicting accounts on whether there is a hop-on hop-off bus that will go to this route. Can anyone tell me if they are still operating?

Margaret, as far as I know, this is the only hoho operator which includes part of the Appia Antica. Their purple route to attractions in that area is only included with their deluxe (3-day) or premium (2-day) ticket levels.

It does not include the aqueduct park. Archeobus hohos used to service the Appia but they went out of business quite a number of years ago.
The Aquaduct Park is a short walk from the metro station Cinecittà (here is a map).

FWIW, the hoho buses in Rome are vastly overpriced for what you (don't) get. You're often better off taking public transit on your own (or even a taxi).

I'll agree with Michael's assessment of Rome's hoho buses for almost ALL of Rome. The Appia might be an exception but yes, there is public transit which services parts of the park:
