I have a 19-day itinerary planned (family of 4, kids are college-age): flying into Amsterdam for a few days, to Munich (by plane), then to Lyon (by air from Munich) and Provence (renting a car) and ending with 7 days in Italy. The initial plan was a couple days in Lake Como, 2 in Florence, and 3 in Rome. Might consider the Cinque Terra instead of Lake Como. Either way, it seems like a long train/bus trip from Provence, we may get have to book a third plane flight. Any suggestions are appreciated.

Given the transit time required to Italy (most of a day) I'd eliminate either Lake Como or Florence and split those 2 days between the other places. It's helpful to count nights, rather than days in each place, since a good part of some days will be spent travelling between places. And don't forget to give serious attention to the total amount of travel time required each time you change locations- hotel to hotel, not just flight or train times. As for CT vs Lake Como- it depends on what your interests are. If your family is into serious hiking and don't mind heat and crowds, then CT may be your choice. Como may be a bit cooler and quieter, but it's all a matter of degree.

You’ve got a lot of moving around already-each location change eats up at least half a day, probably more in your case as you are covering a lot of ground.
Do you have departure flight booked yet? From where?
I’d probably limit to just 2 locations in Italy
Next year is the Jubilee so keep that in mind, expect big crowds especially in Rome.
I would choose Lake Como over the Cinque Terre. Both are crowded but the Cinque Terre unbearably so.
I will “second” what Suki said about Cinque Terre—- way too to crowded to be enjoyable.
If you choose Lake Como, you could drop the car in France on the train line into Italy—-Nice, Menton, or other city—and continue by train to Milan, changing there to a train going to Varenna, one hour away. That would shorten the train journey into Italy.
From Varenna, it is easy to use the train to reach Florence and Rome, with a flight home from there.
However you do this, it is a lot of moving around.
Thanks for all the replies so far. In answer to your questions, this is from mid-June to around the 4th of July. The flight to Amsterdam has already been booked. We definitely want to go through Italy, though maybe Rome could wait for another trip with the Jubilee next year (I've already been there). I thought we'd end there because it's a good departure city.
In Provence I've booked a small villa for 4 nights just outside St. Remy (we could possibly change the dates). That and Amsterdam are the only accommodations I've booked so far. It is a lot of moving around. Maybe from Munich through the Alps to Como and Florence/Siena, skip Rome and fly back from France?
I would drop Italia. And we love Italia. We are planning on 23 days in Italia in 2025.
Lyon is worth 5 days in my opinion. We enjoyed our 5 days there.

We chose to visit Lugano (Lago di Lugano) rather than Lake Como, though I took a day trip to the latter. It's beautiful, lodgings are less costly and it's much less crowded. Food's exorbitant, though.
It's very easy to catch a quick train from Lugano to the Milan Malpensa airport, or to Rome. Worth considering.
Follow-up: thank you for all the suggestions. We decided to limit our Italy destinations to Turin (which I've read a lot of good things about) and Florence.
