We will be taking the train from Venice to Cinque Terre. We will be in Cinque Terre for 3 nights. We will be staying in Monterosso. When we leave, we would like to rent 2 cars (7 ppl) and rent it for 4 days for Tuscany and then drive down to Sorrento and turn it in there. Any recommendations?
Take the train back at least as far as La Spezia, possibly even Firenze. You really don't want a car anywhere close to the CT. Mine got scraped between a box truck and a rock wall on the road to Vernazza. Just glad we were going up, not down, or we would have been pushed off the road and down the mountain!
Yes. Take the train to La Spezia and rent there. Or Pisa airport ( compare prices). According to the AutoEurope website, there are no rental offices in Monterosso. Pisa airport may give you the most choices in terms of pickup hours.

As the others have mentioned, La Spezia is your best bet as it's the closest location with rental agencies. There are no car rental firms in Monterosso (at least not that I've ever seen). The train journey from Monterosso to La Spezia is very quick. As you'll likely be travelling on Regionale trains on that route, DON'T forget to validate your tickets prior to boarding the train on the day of travel.
Note that each driver will require the compulsory International Driver's Permit, which you can easily obtain at any AAA office for a small fee. You'll also need to be vigilant to avoid the dreaded Zona Traffico Limitato traffic areas (ie: hefty fines), which are becoming increasingly prevalent in many towns in Italy.
