This kind of question, requiring very specific familiarity with the exact areas, with bus schedules, and on the holiday in question--if it can be answered at all--is probably best posted at this website, picking one of the forums that matches your itinerary

The trains should run fine but you can bet that the inter-city buses will be on what is known as "Festival" schedule. This usually indicates a reduced schedule. Where are you intending to travel? Italy has a very large quantity of private bus systems and they all have their different schedules. Tuscany has about a dozen bus companies alone.
