We've booked a flight from Venice to Puglia. It will make one stop in Rome. Leaves Venice 6:50 am, leaves Rome 9:05, arrives Bari 10:10.Does anyone know if we will have to de/plane in Rome? Thought I would ask here before attempting to contact Alitalia. Thanks.

Since it is on the ground for 1 hour 10 minutes and they have different flight numbers I think it is safe to say you will be disembarking at the very least and likely changing planes.
