My husband and I are planning a 3 week Italy trip in April 2018 and wanted to spend 4 nights around Florence and the Tuscany Region. We are interested in staying at one of the Agriturismo properties. We will have a car but really want to stay in the Tuscany region but not sure what the best little towns are to be closes to, does anyone have experience in this area, and can you recommend an agriturismo property?

We recently spend three nights at the Villa Palagetto near San Gimignano (http://www.agriturismoniccolai.it/index.html) and had a wonderful experience there. We dined in a delightful patio setting with terrific, reasonably priced food (excellent unlimited wine service from the house vineyard), bucolic countryside scenes right out of a painting, and San Gimignano is within walking distance. Volterra, Siena and Florence are just short drives away. Delightful, friendly hosts. Located in the center of Tuscany, access to the road system is quite easy. Their 100% Syrah wine was so good that we purchased and shipped some home after an enjoyable wine tasting event.
Let me echo Scott's post. We stayed at Villa Arnilu, which is one of the properties run by Agriturismo Niccolai. We were a family of 12 and really enjoyed the villa. Their onsite wine tasting was a hit and we really enjoyed the Tuscan Vernaccia wine, which you can't get everywhere.
Wow, those Niccolai properties look really nice! Someone else recently asked for Tuscany agriturismos and got several more responses. I highly recommend the Ag. Marciano just outside of Siena. A few other folks on this forum enjoyed it as well.

This is in the midlle of what many considers as the most scenic part of Tuscany, the crete senesi.
Very close to Buonconvento, 15 minutes from Montalcino.
We stayed there 6 days in june, it was the most perfect tuscan dream you could imagine.
Reviews aren't unanimous for nothing, the owners are fantastic, genuinely nice people with a big heart. Pictures of the agriturismo talk for themselves but I can tell you it's even better when you are there.
I hope you find the best agriturismo that suits the best your needs and budget.

There are many great agriturismos and you cannot go wrong with all of the ones mentioned above.
Take a look at Cretaiole and Borgo Argenina. Isabella and Elena each cook with their guests one night and truly treat their guests with special treatment - their home cooked breakfasts are a treat !
What is your itinerary ? That will determine the best responses to WHERE in Tuscany to stay. Will you stay in Florence too ? Were you planning on making a day trip into Florence ? Where are you planning to arrive from and head to after this region ?
There are SO many great little villages AND larger towns to visit ! Give us a bit of your plans and you will receive some awesome responses.
We have stayed at several agriturismi in Tuscany but our favorite so far has been IL Rosolaccio between Florence & Siena near San Gimingnano. Our family of 4 stayed a week in apt. "Ginestra" that was 2 bedrooms, 1 bathroom. www.rosolaccio.com They have a beautiful pool also although in April probably won't be open yet. Would stay here again. Buon Viaggio!
