After spending 4 days in Amsterdam at the end of June, we have 8 more days and we think we might want to drive the Romantic Road in Germany with a few stops, then go to Switzerland. Is driving the best way to go? Will car rental be a problem? What are some must sees? Thank you very much for your help!

Renting a car in Amsterdam, returning in Switzerland not a good idea. Too much driving and dropping off car in diff country super expensive.
Would be much easier and cheaper to do some combination of train/car.

Renting a car in one country and returning it in another is quite expensive, especially given the quality and frequency of the trains/flights.

Car Journey - take a train to Koln or Koblenz first and pick up a car, Then hit the high spots on the RS Germany guidebook south to the German-Swiss border, drop the car and take a train in Switzerland.
Train journey - Maybe Day 5 Amsterdam to Koblenz by fast train, and then maybe Middle Rhine Valley (castles and several hour Rhine cruise), 3 hour train trip train to Nuremberg (which is also worth look), and from there it is less than an hour by local train from beautiful Wurzburg and Bamberg and, if you rent a car an hour to Rothenburg ob der Tauber. It is an hour by train from Nuremberg to Munich (Museums, Dachau, Beer gardens, Bus tour of Neuschwanstein or Linderhof from Munich for a day).
Given the frequency of flights (many), and the duration (1 hour) Munich to Zurich, if you want to include Switzerland in the trip I would suggest flying to Zurich and going from there.
