I will be in Rome this year over Easter with my niece. I've been to Rome a number of times previously including the week before and the week after holiday but never during. Anything special we should be aware of in planning our time.
I saw the Vatican museum is closed on the holiday and the Monday after. Is there anything notable open on Sunday we should plan for that day?
Easter is the holiest day of the year of the Christian calendar. As you'll be in Rome, the "anything special" and "anything notable open on Sunday" question can be answered by encouraging you to attend an Easter mass and experience the immense beauty of the service. Do some research and find a church or cathedral with a rich choral heritage for a truly sublime experience.

I'm not sure whether you want to know what else might be open to visit on Easter Sunday, or if there's anything special going on (aside from church services) because of the holiday?
Assuming you're looking for what's open, this upcoming Easter is interesting because it also falls on the first Sunday in April. First Sundays of the month are when the Italian National Museums are free for visiting so that the two coincide could create some challenges!
For instance, the Colosseum/Forum/Palatine are open BUT they fall under National Museum designation so would be free and utterly mobbed. Galleria Borghese, however, while also open and free to visitors on first Sundays, still requires advance reservations to maintain its usual strict crowd control. That might be a good choice?
That "Free Sunday" and Easter Sunday fall on the same day is an unusual enough occurrence that it wouldn't surprise me if the city comes up with some create workarounds but it's too early to tell. Anyway, is this the sort of info you're looking for?
