What to pack for Italy in March? Aver. high is 49 and aver low is 45. We live in WI so anything above 35 will seem balmy at first. What to layer under an REI waterproof/breathable shell? Will I need long underwear under my polyester/nylon pants? We are going from South (Rome) to Florence, Venice, Cinque Terre, etc. Will long sleeve shirts be enough? Which is best, wheeled suitcase or backpack? We are traveling by train & car.

I do have a travel backpack that I used in Spain. However, it's easier to find stuff in a suitcase! But the backpack is probably a better bet since I don't know if all the places we will stay have an elevator.

I have a wheeeled type bag that converts to a backpack.It's perfect.
Bring layers! March can be very chilly (as in the US too I guess) very unpredictable...year before last, we had a freaky March "blizzard" (ok... a little bit of snow...but blizzard for So. Italy!)/ Long underwear though? no. a coat...yes! Italians will still be in winter-wear in March...coats, scarves, gloves, etc.
