EuroStar Italia trains go directly from Venezia Santa Lucia to Roma Termini in 4hr36min. There are other choices, but they require changes and are also slower. See the timetables at trenitalia.com (Italian national rail) or bahn.de (German national rail which includes most of Europe). The Trenitalia site also gives the fares.

Tren Italia's website won't let me pay. I get error messages.
Not a good way to run a business.
I can try calling, though. I need to get from Venice to Rome as well.

The Trenitalia site gives lots of people fits. If you're going to be in Venice a few days before going to Rome, just buy your ticket when you get there. From what I've read, the automatic ticket machines are fast, easy to use, and instructions are avialable in English.
