Joy, we haven't been there yet, but we're gearing up for it. Check out Rick's Italy book, or any other good guidebook. Your local public library probably has a decent selection of guidebooks.
Right now we're looking at seeing daVinci's Last Supper, the Pinacoteca Ambrosia, the magnificent Duomo, the Galleria Vittorio Emanuele II, the Gallerie d'Italia, La Scala Opera House, Bagatti Valsecchi Museum, the Brera Art Gallery, Museo del Novecento, Museo del Risorgimento, and as many churches as we can. Whew! Throw in trattorias, restaurants, markets, piazzas - we figure we can fill up four days without trying too hard.
When are you going?
Consider a side trip to the very atmospheric hill town of Bergamo, roughly an hour away by train, then you take the little funicular up the hill.
The Verona - Vicenza - Padua triumvirate is 90 minutes to a bit over 2 hours away by train.

We loved Bergamo but if your three days includes arrival day, you can easily fill your time just in Milan.
In addition to what's been suggested, San Lorenzo Maggiore and the Basilica of Sant’Ambrogio are musts!
Yes, definitely read Milan section of RS Italy book, major sights described very well, prices, directions, etc, map diagrams, info on using the metro, areas for fun nightlife and more. It's an easy way to plan a self-guided tour of whatever interests you most. I did a Best of Milan walking tour in September with Walks of Italy, primarily because it was the only means of getting Last Supper tickets when I needed them. But the walking tour covered many of the places in RS book. They don't do February tours otherwise I would highly recommend it because their tours are terrific. And another definitely yes: the Duomo!! Totally amazing. If not too cold get the tickets that let you walk the roof. Wow!
I enjoyed La Scala - besides seeing the auditorium there's a museum. The best part of the Duomo is the roof, so much fun to wander around and see the carvings up close. If you like museums, go to Sforza Castle, there are several - plus the castle (fortress) itself.
