2 nights in Rome how to make the most of them

Need suggestions of where to stay - what area. And should we pay for tours and if so recommendations please. Or should we just buy tickets on line and do it ourselves. We arrive Palm Sunday and fly out Tuesday Morning after seeing the Amalfi coast ,Tuscany and hopefully Venice.

Lydia, is the Amalfi Coast, etc., your itinerary before Rome?

You should check opening hours for what you might want to see - most museums and archeological sites are closed on Mondays. The Vatican Museums should be open - I would get tickets ahead of time to skip the lines. It will be Holy Week, a busy time in Rome.

As for where to stay, do you want to be close to the train station or close to the main historical sights? Many posters here like the area around Termini, others (like me) prefer areas near Piazza Navona, the Pantheon, or Trastevere. Either way you will have some commuting (not long in time or distance) to/from Termini and the places you want to see.

Yes we will be in the Amalfi coast March 18-22. Then we travel to Rome for one night to meet friends who are doing the marathon. Then we head with them to Florence Monday until Friday. We have that all figured out. Friday and Saturday night we hope to do Venice. Then Rome Sunday until we leave on Tuesday morning. I think we would prefer to be in the historic area rather than near the train. I will check to see what is open thanks for that tip

The Roma Pass is an outstanding value, with free admission to your first two sights, then discounted admission for the rest, plus it includes your bus/tram/metro costs, but none of the Vatican sights are included. The shortest-duration Roma Pass available is 48 hours, so a Sunday/Monday visit might not make the Pass worthwhile. Cap off your visit, though, with one last Italian gelato!
