I have been searching for a small group tour between Rome and Florence which would allow 2 days/ 1 night to visit a number of towns in Umbria/Tuscany - a ion from Todi, Assisi, Montepulciano, Pienza, Cortona etc. I haven't had any luck in finding anyone offering this type of tour/transfer. I know that there are a number of day tours on offer from Florence, but they seem to either rush their visits to these towns - I'm hoping for a slightly more relaxed pace. I'm not considering car rental so may have to choose only 1 or 2 towns and go by bus/train.

It's hard to see how you'd see a "number" of towns in two days, given travel times, etc. These towns are not particularly well suited to having a bus deliver you from one site to another... most are better suited for wandering on foot. I'd recommend picking one town for a stop between Rome and Florence, at most two... given that you only have two days. I really like Assisi and it's an easy stop by train between Rome and Florence, but your interests may be quite different from mine. Orvieto is also lovely, but you can easily do it as a day trip from Rome; it's about an hour by train. However, you could probably contact one of the tour companies or private guides in Rome or Florence and ask them to plan something for you. It would be costly, but could be done.

When I said a few towns, I was hoping for something like: Day 1 Morning to Todi Afternoon and overnight Assisi
Day 2 Morning to Montepulciano Afternoon in Pienza and onto Florence by evening
