I'd like to take a 2-3 day train trip from Rome with just a small bag. Where can I store my larger luggage? Will the hotels near the Rome terminal do that?

The station in Rome is probably your best bet to get a locker for 2-3 days, assuming you will be heading back that way.

If going and coming from the same hotel, they will probably keep it for you. Most hotels have a left luggage area. Often the security is not real tight but we have never had an problems. Termini has large, staffed, left luggage area on the lower level, right side as you face the tracks. Have no idea about long term storage. Most luggage lockers have a 24 hour limit so that will not work if going for more than a day.
