I know there is a tight train connection to airport for 11:30 plane from Assisi. There will be 7 of us traveling and each with one bag. Is there a van service that would take us directly to be sure we make the plane?

Plane to where? If to US you need to be at airport at about 9am.
You should spend your last night in Rome and have a nice night sleep instead of having to get up at 4:30 in Assisi.

Ask at your hotel. I've used a van before but for the life of me I can't remember the name of the man who runs the taxi service. It'll cost you about 400euro.
Train the night before and a hotel near the airport is really your best bet.
