My husband and I are going to take a cruise that leaves out of Venice but before that we will be in Rome for 2 nights. We want to see the Vatican and Colosium and we love to eat. Where should we stay? A swimming pool would really be nice too. I am hoping for a room no more expensive than 250 a night tops.

When you say you want to see the Vatican do you mean just St. Peter’s or the Vatican Museum and Sistine Chapel? If the latter you might want a hotel close to the Vatican so you can do one of the early morning tours. On a RS Rome tour some years ago we stayed at the Hotel Alimandi Tunisi a couple of minutes walk from the entrance to the Vatican Museum. There is another Alimandi hotel, Vaticano, even closer to the entrance. Line-ups to get in to the Museum begin around 7 am so you should purchase tickets before you leave the States - lots of people on the Forum recommend the Pristine Sistine tour.

If you just want to visit St. Peter’s you could think about staying in Trastevere. Lovely area with lots of restaurants and cafes and easy to get to St. Peter’s and the Colosseum. Not sure what hotels are in that area now as it was just beginning to become trendy when I was there years ago.
We have stayed at the Vaticano 3 times and love it.

Yes, do include the month and year you will be traveling and your maximum nightly budget. Do know that hotel pools aren't as common in Rome, and with only 2 nights (which translates to only 1 full day to explore Rome) you'd have little time to use one anyway.
And while not part of your question, do make sure that your cruise is actually departing from Venice? Some of the bigger ships have been moved to ports in Ravenna and Trieste; that would be noted in your cruise embarkation info. I'd also strongly recommend staying the night before in the city your ship will depart from.

Not completely but all but the smallest ships will be docking away from the island. As mentioned, some lines are moving port to Trieste and Ravenna, and others to the Venetian mainland industrial port of Marghera. They're no longer allowed passage through the Giudecca Canal.
We always stay down the street from the Vatican and take the Underground to the Ottaviano/San Pietro stop. There are a bunch of small hotels and accommodations in the area, and we're familiar with the neighborhood. It's very close to the Vatican Museum.
But if I was just there for two days, I'd be looking for accommodations in the Pantheon neighborhood. It's a very nice area, and the restaurants are very good close by. You're within walking distance of the Tevi Fountains and the Vatican is not far. And the Pantheon is a must see.
Most center city hotels in Rome don't have pools--and you will be too busy to use one anyway.

Rome for two nights? that is like going to Disneyland for one ride. If you have not booked your flights and can make Rome at least 5 nights please do so. If you can not add time to Rome skip it, just spend those nights in Venice. I know your asking for places to stay so here goes. Find a place near the metro or near a taxi stand, so you can get around quickly. You don't have time for a pool or anything else. J
Since you are there for a short period of time I HIGHLY recommend a food tour: lets you try a lot of different things in a short period of time. We really enjoyed the Twilight Trastevere Tour. A good way to spend your first night in Rome since you are out walking around getting the lay of the land.
