I’ll be spending 3 night in Ireland in September. We are planning to take a day trip to Southern Ireland. Is it feasible to visit Waterford and Cork in a single day? I would love recommendations for recommended tours/tour guides as well.

If you're only going to be in Ireland for 3 nights, you'd be spending a vastly disproportionate amount of time traveling from Dublin all the way to Waterford & Cork and back, if it's even humanly possible in one day.
There's plenty to see and do in Dublin. If you really don't care for cities, then choose a place nearer to Dublin where you can settle in for 3 nights and enjoy the countryside. Or, if you are really only interested in the south, get a flight into Shannon instead of Dublin. From Shannon to Cork and back wouldn't be as bad.
