Bus or train. A google search will give you the details.
That will be a long slog, between the ferry and the cross-country rail/bus trip. I'd consider flying, especially if you are actually starting from Rome or Naples or could arrange to do so. You can check skyscanner.com for flights.
It's been over a decade, but four of us took a taxi from Patras to Athens. It is one of my more unusual memories, and totally worth it. But it might not be the best price break for you. It was a spur of the moment decision, but we did negotiate a price before we decided to do it.
Is Brindisi your actual place you will be, or as someone mentioned will it be Rome, Naples, or another place? If you are coming from someplace else, look at flights, if Brindisi, look at flights. Even Brindisi has cheap flights with Ryanair, Alitalia, or others (though Alitalia might be a bit worrisome).
If by ferry, plan on a long overnight, probably sleepless (unless you spring for a cabin) trip, then a several hour journey to Athens. For that I would do bus. Overall the cost, time, and hassle of the ferry and bus is more than a flight.
