Depends on the time of year, your preferences, physical limitations, age and budget. Check "Things to do" just below the green strip on the Santorini forum webpage at www.tripadvisor.com

You should read some guide books to help give you some ideas to then ask about. You could do a day trip to another island, take a catamaran tour, visit ruins, go to the beach, walk around Thera, etc etc.
