En route from Brussels to Lesvos and find ourselves, potentially, with a 12 hour (total time on the ground) layover in Athens. Should we hire a driver and take a tour of the city? Is that feasible? TIA!

A driver (not a guide) at €60 per hour as mentioned in the link above can be an option, but you can also take a taxi (45€) or the express bus X96 (5.5€) or the metro (9€) to the city center.
You will be able to see (quickly) a lot of things on foot by walking through the city center which is largely pedestrianized. and even possibly visit the Acropolis and/or its museum.
At least 2 1/2 hours before your flight to Lesbos, leave Athens to return to the airport
Which 12 hours of the day?
9AM to 9PM is not the same thing as 7PM to 7AM
What about baggage?
