The tourist office offered a good walking tour in 2015. I don't know whether it was daily, and in any case the timing may have changed.

I liked the Historic Green Vault (precious objects that are gorgeous or gaudy, depending on your point of view); it requires a timed ticket. The regular Green Vault is also impressive. Google Dresden Museums to learn about the many other options. In additiom to art, I remember armor, porcelain, and I think there's a Turkish Chamber somewhere. You'll have to be selective; you won't be able to see everything on a day-trip from Prague.

The reconstructed old town is very small. If you aren't that interested in the history of the city you can easily explore the old town on your own.
The highlights of the city are the Zwinger and the Frauenkirche (church of our lady) at the opposite ends of the old town, and the museums located in the castle and the Zwinger. My favorite is the armoury with the Turkish Chamber, but most people prefer the Historic Green Vault. The Historic Green Vault costs extra, while the New Green Vault is included in the castle ticket. The New Green Vault is just as good, but not in reconstructed rooms. AFAIK the number of exhibits is even bigger there.

I would recommend to see two museums. One in the morning, one in the afternoon, with sightseeing and a restaurant between the two visits. If you want to see museums in the Zwinger and the castle you need two tickets or a VIP ticket, which includes the Historic Green Vault, otherwise one ticket for the Zwinger or the castle allows you to see all museums located in the building (minus the Historic Green Vault).

In terms of restaurants I would recommend something local. For dinner Freiberger Schankhaus, which serves mostly Saxon dishes. And for coffee and cake - in Germany Saxons are known as "Coffee Saxons" - either Coselpalais or Dresdner Kaffeestübchen. Coselpalais is in a grand palace, Kaffestübchen is a tiny cafe. Kaffeestübchen offers a great Eierschecke, a local specialty.
