Our DB ticket from Freiburg Hbf shows a transfer at Mannheim Hbf to a bus to take us to Frankfurt airport. Does anyone know where the buses are located and is 20 minutes enough time to transfer from the train to the bus? Thanks for your help.

What kind of a ticket do you have? Is it a train specific, discounted Sparpreis (Savings Fare) Ticket, or a full price fully flexible ticket. Ersatzverkehr is a substitute bus for a train that is not running, probably because or track maintenance. Only about half of the Long Distance routes from Freiburg to FRA use the Ersatzverkehr bus; others are all rail to the airport. If you have a flexible, full price ticket, you could use any one of the other routes and avoid the bus.
If you have a train specific ticket, you have to use the bus. Go out the main entrance to the Mannheim station, turn right and walk parallel to the station about 100 yards to Heinrich Von Stephen Strasse, then another 100 yards along the Strasse to the Ersatzverkehr Bhf. Look it up on Google Maps.
The line from Frankfurt to Mainz is under construction. You have to take the bus. It's been that way for 2+ months now.
The DB info window can help you if you get turned around. It's just inside the main doors.
You can detour by going to Enkenbach and taking the RE65 to Bingen and from there to FRA, but that takes longer.
