I'm starting my travel in Frankfurt , and I think that I'd like to see Trier before heading to Freiburg (which is the primary purpose of my trip.) Is the train the best way to get from Trier to Freiburg ? Rental car ? Car service?

best is in the eye of the beholder...
Train is easy and simple, and as fast as anything else... with some planning it can be quite inexpensive too.
If you want to drive to many places on the way you could rent a car.
I can't begin to imagine how expensive a driver to take you all the way to nearly the Swiss border would cost...
You can pick what fits your budget and needs, wants and desires.
