Traveling entirely by rail our plan is to start in Vienna, go to Salzburg , Munich and end in Berlin (where we have family).
My husband would like to see the castles in Fussen. Is it better to do it as a day trip from Munich? Or stop in Fussen for a night.
Rome2Rio shows the train going from Salzburg through Munich to get to Fussen.
Any guidance would be appreciated.

For my upcoming trip, I decided to spend one night in Fussen rather than do a day trip from Munich for a few reasons. One, I'm driving from Rothenberg, so it's less travel time for the total trip. Two, I read on this forum getting to the castle via either train or tour bus was a hassle. Three, after seeing how much there is to do around Fussen, I decided I'd rather spend 24 hours in the area instead of 8-10.
Bonus: On my way back to Munich, we'll stop in Oberammergau for the alpine coaster.
For you, coming from Salzburg and having to go through Munich to get to Fussen, which is a long train ride tbw, I'd say just spend the night.
Salzburg > Munich > Füssen is the correct routing.
There are other more worthwhile destinations than the "castles" of Füssen which are closer to Salzburg... Hohenwerfen Castle (a true medieval fortress) in Werfen, for example.

And right in between Munich and Salzburg is Herrenchiemsee. Only 1 hour from either Salzburg or Munich to the town of Prien:
A day trip from Munich to Neuschwanstein requires nearly 5 hours round trip on trains and buses. Not very worthwhile for a tour of less than 30 minutes.

A night in Fussen would be my choice too. It’s a quiet, comfortable town and is a good overnight stop. Neuschwanstein, as organized as it is, is the sort of mass tourist stop that can leave you a bit drained. It was nice to get some air, bike around and relax in town afterwards. Sounds like you intend on visiting both castles. Hohenschwangau gets passed by a lot of tour groups, and is for a lot folks (like myself) more satisfying than Neuschwanstein. I traveled to Fussen from Salzburg as well, and while I can’t recall the route I remember it being very simple and easy.
