I will be traveling from Rotenburg ob der Tauber to Rome Italy (not sure of exact date yet). Can anyone tell me about how long it will take? Is there a night train so I don't lose a day traveling? Thanks for your help!

There is a night train from Munich to Rome (CNL 485, lv Munich, 9:08 PM, Ar Rome, 9:22 AM), but you'll have to get from Rothenburg to Munich by day train(s) There is a train leaving Rothenburg at 5:06 PM, Arriving Munich Hbf at 8:39 PM, with changes in Steinach and Treuchtlingen. You'll have 29 minutes in Munich.

More generally, an overnight train only saves you from "using a day" if you can actually sleep on it. The movement and noise of the train stops some people from sleeping altogether. But if you know you can usually fall asleep despite noise and disturbance, go for it.
I've spent 10 nights on night trains, 9 on trains between the west coast and NY when I was in college, 1 in Europe in the 80s. Sleeping was not a problem in my own compartment. Sleeping lying down on a train is far easier for me than in an upright seat on an airplane.
Sleeping on Amtrak in compartment -- lovely! Sleeping in a 6-person couchette compartment between countries not so lovely.
After looking at our assigned surroundings in a 6 person couchette compartment on a night train from Paris to Copenhagen, we opted to sleep in our 1st class seat in a cabin configuration. (had dual reservations just in case ...) Have also taken the Munich to Rome night train in a 2 person sleeping compartment. It was OK, probably got 4-5 hours of light sleep. Looong line at the taxi stand when we arrived in Rome... Have a good trip!!!
