We will be based in Potsdam for 3 weeks beginning June 4. We want to take a 3 or 4 day tour along the Romantic Road. In particular, we want to visit Rothenburg and the castles. We don't want to drive, so would appreciate a recommendation for a tour company.

I don't know if there are any tour companies operating there. There was a similar question about a month ago and no one mentioned one. There is the Romantic Road coach that Rick mentions, but they just blow down the RR without stopping anyplace long enough to really see it, and a lot of place no time at all.
In 2007, I did my own tour of the Road using just public transportation. I spent four full days and two half days on either end, spending as little as two hours in a single day getting from place to place and most of my time in towns along the way.

Potsdam is way up north and Rothenburg is way down south, so a 3-4 day tour would involve you spending quite a few hours on a train just to get to this area.
Nearer to Potsdam are towns like Quedlinburg or Lübeck.

You have beautiful towns and scenery near to you in Saxony as well as castles. The Erzgebirge Mountains have towns like Seiffen where nutcrackers, smokers, and pyramids are made. Goerlitz might be a good stop too.
