We have an extra day in Fussen. We are thinking of going on a day trip (we have a rental car). Any other recommendations on where we could go from Fussen?

Füssen to Luzern is not really possible as a day trip, by train or car.
By road, https://www.viamichelin.com/ says 03h17, 286 km each way, assuming no stops.
By rail, https://www.bahn.com/en/view/index.shtml says 7 hours plus. From Füssen you have to go north first, then double back south.
No time to do anything in Luzern.
Füssen backs onto the Alps. Those mountains you see as a backdrop in all the photos of Neuschwanstein are the Alps. If you have a car I would suggest driving south into Austria.
Yes, it is way too far for a day trip. Go over to the Zugspitze.
Switzerland is a wonderful place to visit. But if you're serious about seeing it then work it into your itinerary as a real destination...

Head to Andechs Monastery & Brewery (about 45 miles from Füssen).
We drove from Füssen and stayed in the town of Pocking. The food is great, the beer greater and the environment very social and fun. If you want to see the brewery, we suggest you book before you go. It is very popular and worth it. A walk to the top of the hill is needed to see the monastery but you can take it slow and visit any of the three beer gardens there
You could drive into Austria to the Heiterwanger See, a 30 min drive (and Plansee). Take a walk around the Heiterwanger See a 1.5 hour walk. There is a boat there in summer too. You can go a bit further to Berwang and take the Sessellift Sonnalmbahn (closed on Tuesdays (Famous for the Winkie Mann on webcam) to the top, visit the Heiterwanger Hochalm and walk down a 1.5 hour walk.

Highline 179/Ehrenburg, Weiskirche, Linderhof Palace, Zugspitze, Ettal Monastery, Oberammergau, Garmisch Partenkirchen are all less than an hour drive from Fussen.
@jason Did you go on highland 176? If so how was it?
From Tegernsee, worthwhile stops are Bad Tolz, Benediktbeuren and Murnau. Murnau has a pretty centre and pleasant, easy walking on the moors for your children.
From Fussen, depends how far you are willing to drive. With your children in mind, the Partnach Gorge would be fun, the main loop should not be too strenuous for your toddlers. Oberammergau for lunch, chair lift and alpine coaster here - https://www.kolbensattel.de/en/
