Does anyone know the fastest way to get from Frankfurt to Beaune? Also, I see trips on the Romantic Road going
From Frankfurt south to Munich, but not visa versa? In other words is it possible to start in Munich, stay overnight in Rothenburg and continue on to Frankfurt the next day? Thanks in advance!

Do you think that the B25 which is the road used to link much of the Romantic Road, and A7 autobahn which is right next door are one-way roads?
They aren't - they are normal roads. The B25 and other roads which link up the various towns on the "Romantic Road" - which is just a marketing name used by the towns - is just a normal two lane road used by farmers and cars and trucks, and works perfectly well north to south as well as south to north. The lanes are the same width, and it goes to the same places in both directions.
The Autobahn also goes in both directions.
By the way, neither Frankfurt or Munich are anywhere near the Romantic Road which is between Würzburg and Augsburg/Füssen.
are you driving, wanting to use the Romantic Road bus or taking trains?
We are taking trains. In an older Rick Steves publication it mentioned going from Munich to Frankfurt via Rothenburg on a bus tour, with a stop off at Rothenburg you could do on your own, then hop the same tour bus to Frankfurt. I dont see that option any more?
We would like to take the train from Frankfurt then to Beaune, France, but maybe flight is better?
A train journey will take between 5 and 6 hours. I doubt you would be much faster by plane, as Beaune is not really close to a major airport.
If you can get up reasonably early in Frankfurt you can get to Beaune in exactly 5 hours using just 3 trains. For example:

Which gives you nice easy transfer times of 17 and 28 minutes so you can go snack hunting and coffee hunting at both places, and gets you to Beaune before noon.
The train doesn't go on the Romantic Road route but certain towns along the route are possible by train.
