I'm traveling (2 of us) from Hamburg to Munich on Friday, 8 June 2018. There seems to be different train lines? What's best? We want comfort and speed. Cheers and thanks.

Most service on this line is by InterCityExpress, which is Germany's fastest and probably most comfortable type of train. Some departures are direct while others require a connection. I'd choose the direct departure that best fits your plan. The 10:00 departure currently has some seats available for as low as 38 euros per person (nonrefundable and limited seats available at this price).

There is only really one rail route, but there are many trains. Usually one departure per hour, sometimes two, mostly without changes. The trip takes about 6 hours.
Go to the DB (German Railways) website: https://www.bahn.com/en/view/index.shtml
Enter, From: Hamburg, to: Munich, and a date and a time, and press "Search" and it will list the trains.
This, like all rail websites, will always find the quickest route.

Don't ignore cheap flights as an alternative. Eurowings and Lufthansa both fly this route several times a day and the competition can make the tickets quite cheap. Right now I see Lufthansa is offering direct one way flights for your specific date for less than 90€. Eurowings is offering direct one way flights from 70€.
