The old town is small enough that location doesn't matter. Only you can define "best" for you.
We have stayed at the Altfraenkishe Weinstube which is in Rick's book and liked it both times. The first time we were in the Weinstube itself and had a small room that was just cozy raised to a power. The second time we were in their annex in really nice room that was huge. Nice people who are very helpful.
we had a car and stayed in a hotel( with car parking) just outside the gate to the town. it really is only a small town so doesn't take that long to walk around. when we were there, their festival was on and took up the whole town, so staying just outside the walls was a good option.
We used the Gerberhaus on the south part of the old city inside the walls and found it quite nice and reasonably priced.
We also stayed at Gerberhaus, recommended by Rick. Very charming. Terrific selection for breakfast--buffet but very fresh and constantly replenished. Rooms very comfortable and location right in the cobbled street area was lovely. If you are looking for a 5 star luxury hotel with room service and a workout room + Jacuzzi, you will not be happy. If you are looking for atmosphere, charm, comfort, it is a cozy lodging perfect for imagining that you live in a walled city.
We stayed at the Hotel Zum Breiterle just inside the walls and loved it. We found it on booking.com. Great value, friendly staff, and yummy breakfast included. I would not hesitate to stay there again when I return someday.
Wouldn't stay there. Instead drive 10 minutes to Castle Colmberg and spend the night in a medieval castle.

Staying within the walls of the town adds to the experience. You just step outside into the golden hours of light for photography in the rising and setting sun. RS Guide will aid your choice, as will the town's website. "Best", like beauty, is in the perception of the beholder.
When are you going? I will be staying at the Gasthaus Goldener Griefen in May. If you are going after that I will give you a report.

Yes! Totally agree. Our favorite hotel is the Eisenhut but it's pricy. We've also stayed at the Klosterstuble and Reichs Kuckenmeister. All 3 are right near the town square, Christmas markets, and shopping. Our last trip we stayed just outside the walls at Hotel Mittermeier. Lovely hotel, charming but it was a walk to the 'action'. My husband loved it but I will choose next time to stay inside the walls near the square.
We stayed just inside the walls at Hotel Cafe Uhl. http://www.hotel-uhl.de/ They provided parking just outside the wall. The most common scene shown of Rothenburg shows the hotel. It was a great location. Don't miss the night watchman tour!!
Is there a place within the wall without a lot of stairs (like 10 or less) and parking close by?
