We will be staying overnight in Tours in September. Any recommendations for centrally located accommodations? Any recommendations for sights to see/activities to do?

Tours itself is great, plenty to do, check our the tourist office site, Also, there's the chateaux (so many chateaux...)
Accommodation, try Maison Jules, a chambre d'hote http://www.lamaisonjules.fr - near the cathedral ( the roman centre of town) or l'adresse www.hotel-ladresse.com in the medieval centre. Both are good. Hotel'univers is also good. Most of the other hotels are OK, we have had clients stay at most of them, but these are the three most popular in with our clients.
