Toulouse to Saint-Lary-Soulon and then to Barcelona (car routes?)

In a couple of weeks, My family will be renting a car in Toulouse and driving into the Pyrenees in one day and staying in Saint-Lary-Soulon (staying a few days). The efficient driving route according to michelin is about 2 hours. Is there anything significant to consider another driving route.

After our stay in Saint-Lary-Soulone, we will cross the border into Spain and make our way towards Tossa de Mar. I have looked at the recommended routes, but does any particular route or city/town/landmark in Northern Spain stick out?

Yes, it will be a lot of driving but we are very excited to experience it in a rental car.

After Spain, we will drive back to Toulouse via Narbonne/Carcassonne (coast route) to return the car back in Toulouse,.

I guess the Michelin route takes you via the A64 motorway to exit 16 at Lannemezan.
This is the best route from a travel time point of view. I don't see what other route you could take except to waste time on secondary roads. Note that you will have tolls a few kilometers after Toulouse and at exit 16. Also watch out for speed cameras, there are a few on the A64.

The travel time will depend mainly on the traffic in Toulouse and its surroundings. 2 hours being an average.

By the way, it's Saint-Lary-Soulan (with an A) and not Saint-Lary-Soulon.

In Spain, going to Tossa de Mar, the obvious and essential stop is of course Barcelona, I guess you will stay there at least one night or two.

Regarding the return to France via the coast, Banyuls-sur-Mer and Collioure are (very touristic) towns where you can stop.

Just before these towns, in Cerbere, the first town after the Franco-Spanish border, stop for a few minutes to admire an Art Deco masterpiece: The Hôtel Belvédère du Rayon Vert in the shape of an ocean liner.
