The Eurostar train from London arrives at Paris Gare du Nord. From there to Saint-Suplice is direct on Metro line 4 (magenta color).

Am confused by your second question. As noted by Laura, Eurostar from London arrives at Gare du Nord. Are you trying to get to the hotel that you asked about from GdN? If so, Laura's answer is perfect. Also, I suggest that you get your metro tickets on the train from London to avoid lines at GdN.
