The itinerary mentions Day 4 is a full day in the Loire Valley where the coach will make a morning run to Chateau de Chenonceau, returning to Amboise in the afternoon. This would be April 28 based on the tour we are on. Is it necessary to buy entry tickets to the Chateau ahead of time, or can I buy them on the day of arrival?

Chenonceau is best booked before you visit so you get the entry time you want. It would be embarrassing to arrive only to find the first entry time available is midday. Even worse to arrive and discover it's full.
