If you want to avoid navigating through Paris, you might want to take the RER A to Marne-La Vallee. There is a Hertz and a Thrifty near the station there. It is in the direction of Trier on the way out of town and fairly easy access to the AutoRoute de L'Est.

I'll add the standard warning: where are you planning to drop off this car? Have you got a quote?
International one way car hire usually incurs a large surcharge.
Gare de Lyon is easy to reach by Metro and has pretty quick and easy access to the autoroutes heading east. As I recall, you don't need to use the Peripherique. Several of the rental companies have branches there.
Just rented a car from Gare de Lyon. (Used Auto Europe and through them, Eurocar). Caveats: while leaving the station was not a big problem, returning the car was tricky. All roads are one-way, so getting to the station by car was not easy. We practiced using Google maps. The actual address for the rental agencies is 193 Rue De Bercy. To get there you have to turn off the Quai de la Rapee (along the Seine) and turn right on Rue Van Gogh which turns into 4 separate roads. Take the far right road (it might not seem like the right one, but it is the very far right one) as it is the only one that will turn onto Rue De Bercy. Turn right at Paul's Bakery and take the first parking garage to the right. Then follow the directions to whatever car rental agency you rented from. Bonne chance.
I did lots of research on pick up spots when renting a car in Paris...I needed somewhere that we could easily walk to from a metro stop, but close enough to the ring road so we could make a fast getaway. I used AutoEurope as a consolidator, and Europcar was the company we rented from...my result...the rental pickup on Foch Ave...it is right past the Arc de Triomphe and easy to get to from the metro stop there, and you don't have to go round that crazy roundabout...it was almost a straight shot (well, a few more manageable roundabouts) to the ring road.
We rented on a Sunday morning, so traffic was light (and because it was a Sun, my choices of pickup spots was narrowed a lot). We also had a GPS screw up and ended up over by the Eiffel Tower, but we had been on the right path at the start and it didn't take too long to get to the highway.
I'd suggest picking up the rental car at Les Invalides. The location is also an RER stop and the Air France Les Cars stop. It's super safe and convenient. If you're staying in the 5th arr, it'd be a short cab ride to Les Invalides.
Last time I rented a car, I picked it up there and drove out of Paris via Champs Elysees on a week day. It was not challenging and less dramatic than I thought it would be. I like the location for the availability of cars with automatic shift. The location is much less busy than the ones in the major train stations. I came across several English speaking travelers also picking up their cars at the same time.
Another good location might be Montparnasse as it is supposedly an easy exit out of Paris on the south side. Additionally, it's a train station so availability should not be a problem.

Pick up the car outside Paris on a train route, one that has a car rental. I would never rent a car in any big city in Europe.
While some suggest avoiding picking up a rental car in city central, my preference would be to avoid the trouble of carrying luggage getting on/off train. It takes time/energy/hassle to get on a train to go outside of the city central. To me, it's a lot easier to pick up the car, put all the belonging in the car and drive out. Driving out of Paris, from my experience, is not as challenging as most people think unless one isn't accustomed to driving daily in heavy traffic.
So I'd take into consideration two main things deciding where to pick up the car... how many bags to carry and driving skills/experience.
