We will be in Colmar for 4 days in mid October, using it as a base for exploring Strasbourg and the Route des Vins. I know we will not need a car for a day trip to Strasbourg (we'll take the train). But what about the Route des Vins? Is there frequent bus service in the area between Colmar and Ribeauville, Kayserberg, and Riquewihr?
A car would make the small wine villages more convenient and fun. Colmar is nice tho many people would prefer to stay in one of the half timbered villages tho Colmar is more convient for a day trip to Strasbourg.
You definitely want a car for the Rue des Vins. As for Strasbourg, we followed Rick's driving instructions and they were excellent ... tell you what exit to take, which lot to park in and which tram to take. Made the trip a piece of cake. A thought on Riquewihr: If possible, time your arrival for later in the afternoon as the day trippers get on their buses and leave about 5:00. It is the neatest of the small towns in the area and is easier to see when the crowds are gone. We based out of there for two nights and loved it.
We stayed in Strasbourg and then picked up a rental car when we moved to Riquewehr on the Route des Vins. We explored many of the villages and can’t imagine seeing it all without a car.
I was in the Alsace in April and a car will make your Trips to the Small towns much easier and also for Castle Haut Koinisbourg which is on top of a Hill.

I found it convenient to use my car rental for the villages. We stayed at an accorhotel in Colmar that had a parking garage.
We based our selves in Riquewihr and explored Kayserberg and other viĺlages by foot, then we could both enjoy the wine!
Another vote for a car. We've done this 8-10 times and prefer to stay in Kaysersburg To eat: Kaysersburg's Au Lion d'Or, a sight to see in itself. The restaurant is still operated by its founding family in its original building. It opened for business in 1729, three years before George Washington was born.
Thanks to everyone for their advice. It looks like we'll be renting a car. I suspected this would be the answer because we found a car indispensable in prior trips to rural France. Merci beaucoup!
