What is the best way to get to Aisne-Marne American Cemetery (from Paris) by rental car? Looks like a visit to Château-Thierry American Monument be done in the same day? Also would like a home base hotel recommendation if possible. Thank you.

The cemetery has a website with all the information needed.
As most of the time in rural France a car is the best way to get there

I was at the cemetary a little over a month ago while touring WWI sites. You can do Chateau Thierry the same day. You should also consider Montfaucon.
This is rural driving with narrow roads.
I stayed at the Logis Hôtel le Tulipier in the small town of Vienne-le-Chateau and found it quite nice. I'd use it again. Mind you, the choices in this area are limited.
