My husband and I will be driving between Amboise and Chamonix the most direct route in early September. We will do probably a 1/2 day still in amboise and then head out. I was wanting to overnight somewhere inbetween to break up the drive. Any recommendations on which town to stop in?

You could stop in Cluny and visit the Abbey there!
Nearby Tournus is slightly further off the route but has some excellent hotels (incl Sept Fontaines) and restaurants.

This is a big city, more than a “town,” but have you been to Lyon, arguably the gastronomic capital of a country focused on gastronomy? Besides the food, Lyon has a number of wonderful sights, too. It’s closer to Chamonix than to Amboise, so if getting there isn’t too much of a drive that day, it could allow you to spend a bit of the next day in Lyon, before continuing into the mountains.
