Trip to York and the Lake District

Planning a 6 day trip visiting York and the Lake District. Is that doable?
Do we need a rental car?

If 6 days is what you have, then it is what you have.

You could probably do 6 days in either destination. And you don't need a car in either destination unless you intend to go well off the usual tourist trail in the Lake District.

For instance if you wanted to get to our latest archaeological find- the biggest ever Viking Villa found in the UK- at Tarns near Silloth, or some of the more obscure valleys a car would be very beneficial.

That is all unlikely if you only have 3 days or so in the Lake District. Of course I would say to split it as two days York, one day travel time and 3 days Lake District. There are other permutations.

The transfer from York to wherever you opt to stay in the Lake District by car is about 2.5 to 3 hours depending on route (but you will definitely want to make stops which lengthens the journey, planned or unplanned). By train more like 5 hours but the scenery on the journey (especially via the Settle and Carlisle line) is something to be savoured. One of the most scenic rail lines in the UK.
