Do come back to check on the details as sometimes TFL doesn’t give optimal connections. Especially for those traveling with luggage.
Otherwise it’s black cabs or ride shares. Uber/bolt/freenow. With dynamic pricing it’s difficult to say which will be cheaper but certainly the easiest with service directly to your hotel. If you have a larger group it may not be that much more than public transportation.
BTW it appears that road crosses the Thames river. If you don’t mind sharing the actual hotel name then you may get better replies.
You also need to define BEST, do you mean fastest, cheapest, simplest? And for how many?

If you have a smartphone, I suggest downloading the Citymapper app. It gives choices for your route as well as updates to any issues.
It will also help for journeys during your stay in London. You can then switch cities if you plan to travel to other places.
Tower Bridge Road isn't short so exactly what is best may depend on your actual hotel. It is in a little bit of a hole between various stations so public transport may require a bit of a walk or use of a bus.
DLR takes you right to Tower Gateway. From there a short cab.
Thank you all for the suggestions. Best for me means easiest. I will have a rolling bag and a backpack. I'm a little "old" lady and would like to avoid lifting luggage or dragging it around too much.
We really like that airport and stay within walking distance of Bank Station so we can use the Docklands Light Rail to and from. If your hotel is within walking distance of Tower Gateway that would be simple.
If you name the hotel people or the nearest landmark people would be able to help you better.
