We have reservations to stay at Thornbury Castle for 3 nights and driving around the Cotswolds. I know it’s not centrally located but can we see several of the villages with 2.5 days?
Assuming you have a rental car, most definitely. Castle Comb, Tetbury, Cirencester, Bradford-on-Avon, and Bibury are somewhat close. The more popular villages Bourton on the Water, Stow on the Wold, and the Slaughters are slightly further away, but they are only an hour from the castle.
If you headed west into Wales you’re not that far from Tintern Abbey and Coch and Caerphilly castles.
I can’t speak to public transportation in that area.

If you cross the M48 bridge into Wales, Chepstow is the nearest castle and it is a relatively short drive N up the A466 to see the ruins of Tintern Abbey. Castell Coch & Caerphilly Castle here further west and would take longer to reach.
The Cotswold villages are within relatively easy reach of Thornbury but you need to plan your order of route.
