I’m only in London for one week on the best of London tour. I think I have to break away one afternoon and make it to the Harry Potter Studio Tour (last tour pick up from London leaves at 1:45). Has anyone done this and if, which afternoon is best? There are 3 afternoons/evenings that mention are free/good for theater but still think I’d likely have to miss something to make it happen.
Are you talking about the Warner Brothers Studio Tour? I think that many people feel it is worthwhile (especially if you have kids), but it's an hour outside of London via public transport, so you may want to take that into account. it's also quite a pricey venue—I believe tickets start at $75 and go up from there.
But it's certainly doable, although it would take a good chunk of time out of your day.

There might be someone out there, but keep in mind that most tour goers are couples or singles and aren't traveling with younger children. That doesn't mean they aren't interested in Harry Potter, but I doubt anyone from a tour would forego a whole day to visit the WBST, especially considering the Rick Steves' tour only has 5+ days to see London.
Have you thought about staying on for an extra day or two? Or coming in a day or two early? That would give you more time (and an entire day) to travel up to the Studio Tour, and I think you would enjoy it more without the time factor. Many airlines still allow you to change your ticket without penalty.
If you type Harry Potter Studio into the search box at the top there are lots of reports of a visit there.
Watford Junction is 20 minutes by frequent fast train from London Euston, then the frequent dedicated shuttle bus from outside Watford Junction station takes 15 minutes. So with average luck the entire journey should take 45 to 60 minutes. That is a lot faster than the dedicated tour bus from central London which has to fight the traffic all the way there, irrespective of it's cost.
What that means is that you would expect to be back in London, by train, about 1 hour after leaving the studios. So if you went up late morning (depending on availability on your day) you should still be able to make the theatre that evening.
Which afternoon is best depends totally on availability on your dates. It sells out very fast, even for people who don't take the tour bus from London.
In 2019, I visited Harry Potter Studios from central London for a 3p admission slot. The whole visit including travel time was 5.5 hours (an hour each way travel time plus 3.5 hours at the studio). I took the train from Euston and then the bus, as a previous poster described.
The studio offers admission slots into evenings, so if I’m reading your query correctly — that you would do this in lieu of theatre — then yes it’s doable on a free afternoon/evening.
Get your tickets from the official website asap because they sell out far in advance. For example it looks like August is completely sold out but most dates in September are still available.

If you have a free afternoon and evening, then you can easily have a great, unhurried visit to the Studios. As mentioned, you will want to go to their official website and book your ticket ASAP. Getting there and back via the train from Euston to Watford Junction is very easy, and the last bus from the Studios to the train station will be on their calendar. I think it's 10 pm in Sept. So there's no reason why you couldn't enjoy the tour activities in the morning, grab a bite of lunch, and then head out to the Studios. DH and I are huge Potterheads- we went in September and were there for at least 6 hours. Don't forget to try the butterbeer.

You don’t need to book a tour from London to visit the HP Studio. Book the ticket yourself at the studio site. Take the train from Euston to Watford Junction. At Watford Junction, there is a free shuttle bus to the studios for ticketholders and you can stay as long as you like (or until they close). I had a ticket for late afternoon and was there until about 9pm. While they call it a ”tour”, the visit for the most part is self-guided. There is an intro when you enter the dining hall at your alotted tour time, but after that you go through at your own pace.

I did this on my own; I haven’t taken the RS London tour. The studio is huge. I am not a big HP fan and I spent 4 hours there. A superfan could easily spend 6 or more hours.

Phred, I believe Mardee is referring to Rick Steves tour goers, not Harry Potter tour goers. :)
Definitely get your tickets to the Studio and get there yourself via Euston. The shuttle bus on the other side is free. The last evening shuttles tend to fill up. When we were there in June, we queued and just missed getting on the shuttle and had to wait. I'd get an Uber to Watford Junction and get the train back to London.
If you have limited time, a bus tour is not the best use of it.
