How long do you need there?
For the day we want to go it looks like 16:00 is the first available which I think is 4pm? Is that okay time to go?
Any tours sell tickets and transportation together so we could get in earlier?

Take the train from London Euston station to Watford Junction. From there a shuttle to the Warner Brothers Harry Potter Studios.

My kids are 9 and 7, are in love with HP. And loved disney world! We are seeing the cursed stone while there.
In that case the kids are going to want a lot of time there. When are you travelling there? Tickets go very quickly. I booked our tickets about 4 months in advance and couldn't get earlier than 10 am. We were there for over 6 hours. I think it's open until 10 pm (2200 hrs), but not sure kids that young could hang in that late.
Combo bus/studio tickets with day tour companies are usually available, but you will pay dearly for it. I've seen GoldenTours mentioned in this regard. Not sure how the return journey would work.

I went at 4pm and while that was fine for me, a real fan would probably think that wasn’t enough time. However, if that is the only option you have just go. The lot is huge and there is a lot to see.
Also, there is a place to eat with HP themed food about half way through; there was a long line for that when I was there. I skipped it. That meant I didn’t get to eat dinner until about 10pm when I got back to London. Plan accordingly.
We were there twice. The first time the children were 8 and 12 years old. We booked through Golden Tours, simply because individual tickets on the website were already sold out. We were there at 10am and it was very crowded.
The second time (August 2022) the children were 14 and 17 years old. We arrived individually and had 4.30 pm as our entry time. We were 20 minutes early and were still able to go straight in.
It was less busy than the first time. The only queue was at the souvenir store checkout.
We were there for about 4 hours both times. The afternoon visit is absolutely fine, it's open until 10pm.
